Description

       The Bst DNA polymerase is part of the DNA polymerase from Bacillus stearothermophilus expressed in E. coli, which is obtained through isolation and purification. The enzyme has 5’'-3' DNA polymerase activity, but does not have 5'- 3' exonuclease activity. Compared with other DNA polymerase, it has high thermal stability, chain replacement activity and polymerase activity, so it is more suitable for isothermal amplification, such as loop mediated isothermal PCR (LAMP), and 68℃DNA sequencing (this condition is especially suitable for GC rich templates and nanogram level templates). This product cannot be used for thermal cycle sequencing or conventional PCR.

        The enzyme activity is defined as the amount of enzyme required to mix 10 nmoL dNTP into acid insoluble sediment within 30 minutes at 65℃.

        50 mM KCl
        1 mM DTT
        0.1 mM EDTA
        50% Glycerol
        0.1% Triton® X-100
 
Typical LAMP reaction
 

 

 

Add the following in 0.2ml thin wall reaction vial
        Bst DNA polymerase 2µl
        10×PCR buffer                                      5µl
        100 mM MgSO4                                   3µl
        10 mM dNTPs mix 7µl
        F3 and B3 Primers (Final 200nM)       x µl
       FIC and BIC Primers (Final 800nM)    y µl
       Template DNA 1~10µl
        Add water totally                                  50ul
Gently mix and centrifuge, then put it into a 65℃ water bath to react for 1 hour.
 
Stability

 

   Stable at -20℃for 36 months.
